Description
WHAT IS KETOROLAC?
Ketorolac tromethamine is a racemic mixture of [-]S and [+]R ketorolac tromethamine. Ketorolac tromethamine may exist in three crystal forms. All forms are equally soluble in water. Ketorolac tromethamine has a pKa of 3.5 and an n-octanol/water partition coefficient of 0.26. The molecular weight of ketorolac tromethamine is 376.41. Its molecular formula is C19H24N2O6.
HOW DOES IT WORK?
This medication is a nonsteroidal anti-inflammatory drug (NSAID). It works by blocking your body’s production of certain natural substances that cause inflammation. This effect helps to decrease swelling, pain, or fever.
WHY USE?
Ketorolac is almost entirely bound to plasma proteins (>99%), which results in a small apparent volume of distribution with extensive metabolism by conjugation and excreted via the kidney. The analgesic effect occurs within 30 minutes, with maximum effect between 1 and 2 hours and duration of 4 to 6 hours. Ketorolac demonstrates analgesia well beyond its anti-inflammatory properties, which are between those of indomethacin and naproxen; but ketorolac can provide analgesia 50 imes that of naproxen. Ketorolac has antipyretic effects 20 times that of aspirin and, thus, can mask a febrile response when administered during the postoperative period.
